Once you connect to it, it is mounted on /Volumes and can be accessed as if it was a folder by Subsonic. It is, however, mounted as the user who made the connection, which may not be the same user Subsonic runs as, so it may lack permission to write to the drive for things like cover art.
The trick is making sure it is automatically mounted (as the right user) when the machine boots. This involves adding a line in /etc/fstab I believe, but I've never needed to do this for OSX yet, only Linux. On Linux, my servers access mounted drives from an independent box all the time.
